The Octave Up Down buttons have a dual function that will allow you to transpose the keyboard. Box 1 | Drug response variation among individuals and populations During the past 50 years of pharmacogenetic research42, we have learnt that variation between individuals that is influenced by genes and other factors is relevant to the efficacy of all drugs. We now know that metabolic enzymes are affected not only by SNPs of which the human genome contains more than 10 million ; , but also by other genomic variation, such as gene duplications and deletions, mutations in regulatory genes, and probably by recentlydescribed large-scale copy number variations43, 44. Increasing numbers of relevant polymorphisms are being discovered. Most relevant to our discussion, we also know that the frequencies and distributions of harmful and protective polymorphisms vary greatly between human populations22, 34, 45. Given all of the above, it is valid to study traits that are predominantly expressed in specific populations46. Such studies might provide a molecular basis for population differences in drug-metabolizing enzymes for example, cytochrome P450 REFS 47, 48 ; , sulfotransferases49, 50 and methyltransferases51 ; , transporters such as ABC1 REFS 34, 52 , receptors such as adrenergic receptors3, 47 ; and other factors that are involved in differential drug responses and disease susceptibility. Many of the population-group differences that are documented are likely to have important medical and public-health implications10, 5355 and elidel.

It has been amazing to witness the growth of CAPRISA in staff from just 6 people in January 2003 to the current staff complement of 239. As a result of this rapid growth monthly staff meetings are no longer feasible and have been replaced with bi-annual team building function. The first of these functions was a medieval theme and was organized by the eThekwini site team. At first the staff were apprehensive about having to dress up in costumes, but this soon became a highlight of the day to dress up and let their hair down. The food was plentiful and the whole ambiance of the room got everyone back to the medieval times. Some of the words used to describe the event by our staff members were: awesome, fun, fantastic, amazing, well planned, good company, food and atmosphere, felt like we were a team!!! I would like to thank all staff for making this function a success as it proves that we are a great team when working together. 600 FILARIAL GENOMICS: GENE DISCOVERY AND EXPRESSION. Laney SJ, Williams SA. Smith College; University of Massachusetts, MA. Projects to study the genomes of two filarial parasites, Brugia malayi and Onchocerca volvulus, were initiated in 1995. The ultimate objective of these genome projects is to identify new vaccine candidates and drug targets for lymphatic filariasis and onchocerciasis. The first goal of each genome project was the identification of thousands of new genes. These genes have been identified by randomly selecting clones for DNA sequence analysis ESTs ; from cDNA libraries that have been constructed from many life cycles stages of B. malayi and O. volvulus. To date, over 25, 000 B. malayi ESTs have been entered into the National Center for Biotechnology Informations dbEST database and about 8000 new genes have been identified estimated to be about 40% of the complete set of B. malayi genes ; . About 15, 000 O. volvulus ESTs have been entered into the dbEST database representing about 4, 200 new genes. In addition to new gene discovery, the ESTs can be used to identify genes that are most highly expressed at each stage of parasite development. Such analyses can provide insights into the biology of the organisms and can suggest new molecules for study as drug targets and vaccine candidates. In Figure 3, study 1 and study 2 refer to scans before and after treatment of a patient and can be explained as follows. In study 1, 4 black segments are found to be less than -3 SD strongly abnormal blood flow ; , and 2 orange segments are between 2 SD to -3 abnormal blood flow segments ; . Thus of the 54 segments, 6 segments represent 11% of the total. A decrease in the number of abnormal segments and similarly, from the table an increase in the number of normal segments, occur in this example. These were compared for interprocedural effects as well as for regional effects for a particular drug intervention. The comparisons were assessed for significant differences using Student's two-tailed t-tests at 1% and 5% levels of confidence.
